
Why folinic acid, and why this combination
Research has found that many children with ASD have trouble getting folate into the brain despite normal blood levels, a pattern linked to folate-receptor antibodies (Rossignol & Frye 2021, PMC8622150). Folinic acid can enter by an alternate route that stays open when the main receptor is blocked (Frye 2020, PMC7477301), and in a randomized, placebo-controlled trial children with ASD and language impairment who received it improved on verbal-communication measures, most in the antibody-positive subgroup (Frye 2018, PMC5794882).
Improved adds the cofactors the pathway needs: two active forms of B12, riboflavin-5-phosphate for the MTHFR enzyme, and pyridoxal-5-phosphate for neurotransmitter production.*

We know of no other capsule that combines folinic acid with active B2, active B6 and two forms of B12 in one dose. The high-dose folinic powders on the market are single-ingredient with a few cofactors; children's multivitamins are broad but low-dose; prescription leucovorin is the same folate molecule as a tablet, typically made with lactose.
Speech Essentials is made with folinic acid (calcium folinate), the same folate molecule, 5-formyltetrahydrofolate, that leucovorin is based on and the compound used in the folinic-acid studies in children with ASD (Frye 2018, PMC5794882; Renard 2020).
